I have six girls wanting to share my four bedroom house can I do this under a assured shorthold tenancy or does it become a hmo.
Telephone your local council and ask them. Probably the EHO Dept. I had four students that wanted to share and my local council said it wasn't a HMO but they may differ.

Until the new Housing Act HMO legislation starts - the house may or may not be an HMO depending on how it is occupied and the relationship of the members of the 'Group' to one another.
Most authorities in my area (West Yorkshire) do an assessment asking questions which reflect features in the case law judgements which caused the uncertainty (Sheffield/Barnes, Rogers/Islington).
In two neighbouring identical houses each with 6 occupants - all students - one WAS an HMO and the other wasn't.
If you are a private landlord it WILL be an HMO in the near future and if 3 storeys and above will be subject to mandatory licensing.
Most HMO tenants are on Assured Shorthold tenancies? - I'm not sure what you mean by this?
Others may be able to advise about the numbers of people on a tenancy agreement - but it is usually still an AST - most landlords who let to a group make sure they have joint and several liability to protect them if someone leaves.
